皰疹病毒甲亞科包括單純皰疹病毒屬、水痘病毒屬、馬立克氏病病毒屬、傳染性喉氣管炎病毒屬，主要成員有：人單純皰疹病毒I、II型（HHV-1， HHV-2），馬皰疹病毒I、III型（EHV-1、-3）、豬皰疹病毒I型（SHV-1）、牛皰疹病毒II型（EHV-2）、獼猴皰疹病毒I型（CHV-1）、貓皰疹病毒I型（FHV-1）、人水痘-帶狀皰疹病毒、馬立克氏病毒（MDV，也稱鶉雞皰疹病毒I型）、火雞皰疹病毒（也稱鶉雞皰疹病毒II型）。
皰疹病毒甲亞科成員基因組核苷酸序列或推測的氨基酸序列形成α皰疹病毒亞科各屬成員的特定譜系，保守基因模塊按?皰疹病毒甲亞科的特徵排列。本亞科成員由短獨特序列（Us）和側翼倒置重複（IRs和TRs）組成基因組的一個區域含有在HHV-1基因組中發現的同源基因，並作爲亞科的特徵。在培養細胞中，病毒生產性感染成纖維細胞，並在活體感染上皮細胞，許多成員感染自然宿主引起外腫，通常是空泡上皮病變。
